Abstract
In order to simultaneously determine the accurate content of guanine, thymine, adenine, adenosine and cordycepin in Cordyceps sinenis by RP-HPLC method. An AichromBond-AQ C18 column (250mm x 4.6mm x 5 mu m) is used as the analytical column, and acetonitrile-water (7:93) as the mobile phase, the flow rate is set as 0.8mL/min, the detection wavelength is 254 nm and the column temperature is 30 degree C. The linear ranges of guanine, thymine, adenine, adenosine and cordycepin are 1.01-5.50 mu g/mL(r=0.9995), 0.05-1.01 mu g/mL (r=0.9998), 0.20-1.98 mu g/mL (r=0.999 5), 1.01-5.07 mu g/mL (r=0.9995), 0.10-1.03 mu g/mL (r=0.9996), respectively. The mean recoveries of the five components are 99.1%, 97.8%, 98.6%, 100.4%, 99.2%, respectively. The RSD U=5) is 1.15%, 2.34%, 2.01%, 1.75%, 1.50%, respectively. The method is simple, quick, accurate, reproducible, and can be used to determine the content of guanine, thymine, adenine, adenosine and cordycepin in Cordyceps sinenis at the same time.
