Abstract

We have constructed a recombinant DNA cloning vector with a high strand-separation melting temperature. This feature is the result of an insertion of a short sequence of dG:dC base pairs. The new vector is useful for cloning DNA segments in anticipation of forming R-loops for the purpose of either transcriptional mapping or gene isolation.
